ABSTRACT

Objective: Securidaca inappendiculata is a medicinal plant frequently used in the treatment of inflammatory diseases in south China. In this study, we aimed to explore its bioactive constituent which contributes to the anti-inflammatory activity. Methods: Polyphenol-enriched and polyphenol-deprived fractions (PRF and PDF, respectively) were separated from the ethanolic extract by HPD300 macroporous resin-based method, and their anti-inflammatory activities were investigated on a lipopolysaccharide (LPS)-induced acute lung injury (ALI) model in rats. ABSTRACT

In the present work, the phytochemical screening, polyphenolic content, antibacterial activity and antioxidant activity of Securigera securidacaseeds in methanol were carried out. Phytochemical analysis of seeds showed the presence of alkaloids, flavonoids, saponins, terpenoids, steroids and glycosides. Total phenolic content was estimated by Folin Ciocalteau method and the result showed the highest phenolic content of 62.28 mg/g. Methanolic extract was screened for antibacterial activity by disc diffusion method and it found to be potent. The MIC of methanol extract identified by broth dilution method showed a MIC value of 0.25 mg/ml for both E. coliand Kl. Oxytoca, and also 0.5 mg/ml for both S. aureusand S. epidermis.The antioxidant effect of the seeds was tested by DPPH scavenging activity as in vitro assay. The extract had potent inhibitory activity (IC50) value of 0.057mg/ml. The finding experimental results showed that methanolic extract of Securigera securidacais important as a source of antibacterial activity and polyphenolic antioxidants.

ABSTRACT

Objective To study the chemical constituents from the rhizomes of Securidaca inappendiculata. Methods The concrete exacted by 95% EtOH from the rhizomes of S. inappendiculata was isolated and purified by chromatography on macroporous resin, silica gel, MPLC, gel, preparative HPLC, etc. The structures of the chemical constituents were elucidated by means of physicochemical properties and spectroscopic analysis. Results Five compounds were isolated and identified as 2- methylene-butanoic acid 4-O-[(β-D-glucopyranosyl)oxy]-intramol-1,6’-ester (1), 3-methoxyl-4-O-β-D-glucopyranosyloxy-benzoic acid methyl ester (2), threo-4,7,9,9’-tetrahydroxy-3,3’-dimethoxy-8-O-4’-neolignan-4-O-β-D-glucopyranoside (3), eucomegastigside A (4), and acernikol-4″-O-β-D-glucopyranoside (5). Conclusion Compound 1 is a new hemiterpene glycoside named securiterpenoside D, and compounds 2-4 are isolated from Polygalaceae for the first time.

AIM To study the chemical constituents from Securidaca inappendiculata Hassk..METHODS The dichloromethane and ethyl acetate fractions of S.inappendiculata 95% ethanol extract were isolated and purified by silica,gel column and recrystallization,then the structures of obtained compounds were identified by physicochemical properties and spectral data.RESULTS Ten compounds were isolated and identified as 8-hydroxy-1,3,4-trimethoxyxanthone (1),1,2,7-trimethoxyxanth-one (2),4-hydroxy-3,5-dimethoxybenzaldehyde (3),sesamin (4),2-methoxy-3,4-methylenedioxybenzophenone (5),1,3,7-trihydroxy-4-methoxyxanthone (6),1,3,7-t-rihydroxy-2-methoxyxanthone (7),2-hydroxy-1,7-dimethoxyxanthone (8),3,8-dih-ydroxy-1,4-dimethoxyxanthone (9),oα-spinasterol (10).CONCLUSION Compound 1 is first found in the form of natural product,compounds 2-4 are isolated from genus Securidaca for the first time.

ABSTRACT

Nine compounds, including five lignan glycosides (1-5), three sucrose esters (6-8), and one organic acid ester (9), were isolated from the ethanol extract of the roots of Securidaca inappendiculata by various chromatographic methods including silica gel, MPLC and preparative HPLC. Their structures were elucidated as acernikol-4″-O-β-D-glucopyranoside (1), (7R, 8S)-dihydrodehydrodiconiferyl alcohol 9-O-β-D-glucopyranoside (2), (7R, 8S)-dihydrodehydrodiconiferyl alcohol 4-O-β-D-glucopyranoside (3), (7R, 8S)-dihydrodehydrodiconiferyl alcohol 9'-O-β-D-glucopyranoside (4), (7R, 8S)-5-methoxydihydrodehy-drodiconiferyl alcohol 4-O-β-D-glucopyranoside (5), 3, 6'-O-diferuloylsucrose (6), 3-O-feruloyl-6'-O-sinapoylsucrose (7), sibricose A5 (8), and mehyl ferulate (9) on the basis of 1H-, 13C-NMR and MS experiments. Compounds 1-5, 8, and 9 were isolated from the Securidaca genus for the first time. Compounds 2, 3, and 7 exhibited weak cytotoxic activities against Hela and MCF-7 cell lines.

ABSTRACT

Abstract Securigera securidaca (L.) Degen & Döefl., Fabaceae, has been widely used in the Iranian, Indian and Egyptian folk medicine as antidiabetic and anti-hyperlipidemic remedy. Phenolic profiling of the ethanolic extract (90%) of the flowers of S. securidaca was performed via HPLC-DAD-MS/MS analysis in the positive and negative ion modes. The total polyphenols and flavonoids in the flowers were determined colorimetrically, and the quantification of their components was carried out using HPLC-UV. Total phenolics and flavonoids estimated as gallic acid and rutin equivalents were 82.39 ± 2.79 mg/g and 48.82 ± 1.95 mg/g of the dried powdered flowers, respectively. HPLC-DAD-MS/MS analysis of the extract allowed the identification of 39 flavonoids and eight phenolic acids. Quantitative analysis of some flavonoids and phenolics (mg/100 g powdered flowers) revealed the presence of isoquercetrin (3340 ± 2.1), hesperidin (32.09 ± 2.28), naringin (197.3 ± 30.16), luteolin (10.247 ± 0.594), chlorogenic acid (84.22 ± 2.08), catechin (3.94 ± 0.57) and protocatechuic acid (34.4 ± 0.15), in the extract. Moreover, the acute toxicity, hypoglycemic and hypolipidemic effects of the extract were investigated using alloxan induced diabetes in rats in a dose of 100, 200, and 400 mg/kg bwt. The ethanolic extract was safe up to a dose of 2000 mg/kg. All tested doses of the flower extract showed marked decrease in blood glucose level by 31.78%, 66.41% and 63.8% at 100, 200 and 400 mg/kg bwt, respectively, at p < 0.05. Regarding the anti-hyperlipidemic effect, a dose of 400 mg/kg of the flower extract showed the highest reduction in serum triacylglycerides and total cholesterol levels (68.46% and 51.50%, respectively at p < 0.05). The current study proved the folk use of the flowers of S. securidaca as anti-diabetic and anti-hyperlipidemic agent which could be attributed to its high phenolic content.

ABSTRACT

The aqueous, ethanol and acetone crude extracts and dichloromethane, hexane, ethyl acetate and nbutanol fractions of the Securidaca longepedunculata roots and Vernonia glabra leaves were studied for their antimicrobial activity against Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus and Candida albicans using agar well diffusion method. The phytochemical presents, as well as the minimum inhibitory concentration (MIC) and minimum bacteriacidal/fungicidal concentration (MBC/MFC) of the extracts were also determined using standard methods. Results obtained indicated that Vernonia glabra leaves acetone extracts had excellent antimicrobial activity against E. coli, P. aeruginosa and ethanol extracts against S. aureus. The n-butanol fractions had the best activity against E. coli and S. aureus, dichloromethane fraction against P. aeruginosa and ethyl acetate fraction against C. albicans. For S. longepedunculate root ethanol extracts showed best activity against E. coli, acetone extract against P. aeruginosa and aqueous extract against C. albicans. The n-butanol fractions had best activity against E. coli, P. aeruginosa and C. albicans. These results verified the claims by traditional healers in Malawi that the plants extracts treats bacteria related ailments such as diarrhoea and could be a potential source for development of phytomedicine.
